چکیده

Horse liver alcohol dehydrogenase is a nonspecific enzyme (1). It has been observed by Kaplan and Ciotti (2) that the enzyme catalyzes a transfer of hydrogen from ethanol to pyridine 3-aldehyde. We have found that the enzyme catalyzes a rapid interconversion of lactaldehyde to acetol. The evidence suggests that this isomerization proceeds through an oxidation-reduction process similar to that observed by Kaplan and Ciotti. The present paper describes this reaction.

Aldehyde dehydrogenase, a diphosphopyridine nucleotide-linked enzyme.

The dismutation of acetaldehyde to ethyl alcohol and acetic acid has been shown to occur in animal tissues (1, 2). The dependence of this reaction on diphosphopyridine nucleotide was demonstrated by von Euler and Brunius (3). Partial purification of the responsible enzyme system was achieved by Dixon and Lutwak-Mann (4). These authors clearly demonstrated that the mutase activity was independen...

Effect of Folic Acid and Analogues on the Dehydrogenase and Isomerase Activities of Liver Alcohol Dehydrogenase.

Several enzymes possess more than one specific function which can be selectively altered by various agents. For example, glutamic dehydrogenase oxidizes both alanine and glutamic acid (1) ; enzymic activity toward glutamic acid was inhibited by diethylstilbesterol while that toward alanine was stimulated by the same compound. The present communication describes studies on the alterations of the...

Overview of the role of alcohol dehydrogenase and aldehyde dehydrogenase and their variants in the genesis of alcohol-related pathology.

Alcohol dehydrogenase (ADH) and mitochondrial aldehyde dehydrogenase (ALDH2) are responsible for metabolizing the bulk of ethanol consumed as part of the diet and their activities contribute to the rate of ethanol elimination from the blood. They are expressed at highest levels in liver, but at lower levels in many tissues. This pathway probably evolved as a detoxification mechanism for environ...

Hairpin Ribozyme Genes Curtail Alcohol Drinking: from Rational Design to in vivo Effects in the Rat

Ribozyme genes were designed to reduce voluntary alcohol drinking in a rat model of alcohol dependence. Acetaldehyde generated from alcohol in the liver is metabolized by the mitochondrial aldehyde dehydrogenase (ALDH2) such that diminishing ALDH2 activity leads to the aversive effects of blood acetaldehyde upon alcohol intake. A stepwise approach was followed to design genes encoding ribozymes...

Amino ketone formation and aminopropanol-dehydrogenase activity in rat-liver preparations.

1. Rat tissue homogenates convert dl-1-aminopropan-2-ol into aminoacetone. Liver homogenates have relatively high aminopropanol-dehydrogenase activity compared with kidney, heart, spleen and muscle preparations. 2. Maximum activity of liver homogenates is exhibited at pH9.8. The K(m) for aminopropanol is approx. 15mm, calculated for a single enantiomorph, and the maximum activity is approx. 9mm...
